Triple-Drug Treatment Is Effective for Lymphatic Filariasis Microfilaria Clearance in Samoa

چکیده
Following the first triple-drug mass drug administration (MDA) for lymphatic filariasis in Samoa 2018, unexpected persistence of microfilaria (Mf) positivity 18 (15%) 121 antigen-positive persons was observed a nationwide household survey 1–2 months later. Of Mf positive persons, 14 reported taking MDA, raising concerns about MDA efficacy. In 2019, 5–6 after 2018 survey, monitored treatment study done to evaluate directly weight-based these individuals. presence and density were assessed before 7 days treatment, using 1 mL membrane filtered venous blood, 60 uL thick blood films on slides prepared from or fingerprick blood. All participants still filters pre-treatment samples, but two negative by slide made same samples. cleared completely day 12 13 followed up, 30 remaining participant. Filtered EDTA samples (to reduce clumping Mf) is preferred over alone improving likelihood detecting estimating their density. The strategy effective at clearing when given taken correct dose.

Seroprevalence and Spatial Epidemiology of Lymphatic Filariasis in American Samoa after Successful Mass Drug Administration
BACKGROUND As part of the Global Programme to Eliminate Lymphatic Filariasis (LF), American Samoa conducted mass drug administration (MDA) from 2000-2006, and passed transmission assessment surveys in 2011-2012. We examined the seroprevalence and spatial epidemiology of LF post-MDA to inform strategies for ongoing surveillance and to reduce resurgence risk. METHODS ELISA for LF antigen (Og4C3...
متن کاملMolecular Xenomonitoring Using Mosquitoes to Map Lymphatic Filariasis after Mass Drug Administration in American Samoa
BACKGROUND Mass drug administration (MDA) programs have dramatically reduced lymphatic filariasis (LF) incidence in many areas around the globe, including American Samoa. As infection rates decline and MDA programs end, efficient and sensitive methods for detecting infections are needed to monitor for recrudescence. Molecular methods, collectively termed 'molecular xenomonitoring,' can identify...
متن کاملPrevalence and risk factors associated with lymphatic filariasis in American Samoa after mass drug administration
BACKGROUND In 2000, American Samoa had 16.5% prevalence of lymphatic filariasis (LF) antigenemia. Annual mass drug administration (MDA) was conducted using single-dose albendazole plus diethylcarbamazine from 2000 to 2006. This study presents the results of a 2007 population-based PacELF C-survey in all ages and compares the adult filarial antigenemia results of this survey to those of a subseq...
متن کاملEfficacy, Safety, and Pharmacokinetics of Coadministered Diethylcarbamazine, Albendazole, and Ivermectin for Treatment of Bancroftian Filariasis.
BACKGROUND Available treatments for lymphatic filariasis (LF) are limited in their longterm clearance of microfilaria from the blood. The safety and efficacy of a single-dose triple-drug therapy of the antifilarial drugs diethylcarbamazine (DEC), ivermectin (IVM), and albendazole (ALB) for LF are unknown. METHODS We performed a pilot study to test the efficacy, safety, and pharmacokinetics of...
متن کاملEffectiveness of a triple-drug regimen for global elimination of lymphatic filariasis: a modelling study.
BACKGROUND Lymphatic filariasis is targeted for elimination as a public health problem by 2020. The principal approach used by current programmes is annual mass drug administration with two pairs of drugs with a good safety profile. However, one dose of a triple-drug regimen (ivermectin, diethylcarbamazine, and albendazole) has been shown to clear the transmissible stage of the helminth complet...
